Fahad Nyanja’s late goal gave the Tons the derby spoils at Mile End Stadium.
Nyanja struck seven minutes from time to seal the points for the visitors, following their heartbreaking FA Cup penalty shoot-out defeat at Bowers in midweek, as they moved up to seventh place in the table.
Tower Hamlets boss AJ Olatunbosun will look for his men to bounce back when they travel to Takeley on Tuesday.
London Bari 3 Sporting Bengal 1
Bari got the better of Bengal in another local derby at the Old Spotted Dog ground.
The home side had the better of the first half, but Bengal hit back after the break through Prince Pipin.
The visitors saw Pipin miss from the penalty spot, though, and Bari secured the points to move up to fifth after their second win in three outings.
Newham 3 Hullbridge 2 (at Hullbridge)
Newham finally got their campaign underway at Lower Road, with their Terence McMillan Stadium not ready to stage matches yet.
Hullbridge opened the scoring on seven minutes through Kurtis Clarke, but Jack Sawyer levelled before Newham saw a penalty saved.
Jamie Buffin put Hullbridge 2-1 up before half time and went close to a third after the restart when htting a post.
Ross Quinton levelled on 65 minutes, though, and Charlie Massey-Read netted an 80th-minute for Zakir Hussain’s men.
